摘要:
      摘要 目的:探讨电针百会、大椎穴促进脑缺血大鼠学习记忆功能的可能机制。 方法:将30只雄性SD大鼠随机分为3组,即假手术组,模型组及电针组。采用永久性双侧颈总动脉结扎(permanent bilateral common carotid artery occlusion, BCCAO)模型。电针组造模后第二天开始行百会、大椎穴电针治疗,每天1次,每次20min,共28天。采用Morris水迷宫检测大鼠的学习记忆功能,免疫荧光染色观察海马神经元存活情况,蛋白质印迹法(Western blot,WB)检测海马组织神经生长因子(nerve growth factor,NGF)、酪氨酸蛋白激酶A(tyrosine kinase A, TrkA)及磷酸化酪氨酸激酶A(phosphorylated-tyrosine kinase A, p-TrkA)蛋白的表达情况。 结果:模型组大鼠海马神经元标志物NeuN阳性细胞较假手术组显著减少(P<0.01),电针组海马神经元标志物NeuN阳性细胞较模型组显著增加(P<0.01)。Morris水迷宫结果显示与假手术组相比,模型组大鼠的平均潜伏期明显延长(P<0.01),穿过平台次数显著减少(P<0.01)。与模型组相比,电针组大鼠的平均潜伏期明显缩短(P<0.01),穿过平台次数显著增加(P<0.05)。WB结果显示与假手术组相比,模型组大鼠海马组织NGF和p-TrkA蛋白表达量显著下降(P<0.01),而电针组大鼠海马组织NGF和p-TrkA蛋白表达量比模型组增加(P<0.01),各组间TrkA蛋白表达量没有显著性差异(P>0.05)。 结论:电针可以改善脑缺血大鼠学习记忆功能障碍,电针发挥作用的机制可能与激活NGF/TrkA信号通路有关。

      Abstract Objective:To explore the mechanism of electroacupuncture(EA) in promoting learning and memory of rats with cerebral hypoperfusion. Method:Thirty male SD rats were randomly divided into three groups: the sham group, the model group and the EA group. Permanent bilateral common carotid artery occlusion(BCCAO) model was used in this study. EA was started on the second day after the model surgery, the EA group was given EA therapy at Baihui and Dazhui acupoint for 20min/day , a total of 28 days. Morris water maze was used to assess the learning and memory function of rats. The survival of hippocampal neurons could be determined by immunefluorescence staining and Western blot was used to detect the expression of NGF(nerve growth factor)、TrkA (tyrosine kinase A) and p-TrkA (phosphorylated-tyrosine kinase A) protein. Result: Immunofluorescence staining revealed the marker NeuN in hippcampal neurons was decreased in the model group compared with the sham group(P<0.01), increased in the EA group compared with the model group(P<0.01). Morris water maze test showed that compared with the sham group, the escape latency in the model group was significantly longer(P<0.01), and the times of crossing the platform were reduced(P<0.01). Compared with the model group, the escape latency in the EA group was shortened(P<0.01) and the times of crossing the platform were increased(P<0.05). WB results showed that the expression of NGF and p-TrkA proteins was significantly decreased in the model group(P<0.01) compared with the sham group, and increased in the EA group compared with the model group(P<0.01).There were no significant differences between the three groups in TrkA protein expression(P>0.05). Conclusion:EA can improve the learning and memory dysfunction in cerebral ischemia rats, and the mechanism may be related to the activation of NGF/TrkA signaling pathway.
