摘要:
      目的:探讨不同强度的运动对2型糖尿病大鼠热休克蛋白72(HSP72)基因的表达和心肌凋亡的影响。方法:雄性SD大鼠36只,分成糖尿病组(DFM)和对照组(CFM)。糖尿病造模成功后随机分成糖尿病非运动组(DRN),糖尿病高强度运动组(DRH),糖尿病低强度运动组(DRL);对照组分成正常大鼠非运动组(CRN),正常大鼠高强度运动组(CRH),正常大鼠低强度运动组(CRL)。采用活动平板耐力运动8周,检测大鼠心肌HSP72基因表达以及TUNEL染色观察。结果:糖尿病大鼠心肌HSP72基因表达均显著低于正常大鼠,其中CRL组和DRL组显著高于CRN组和DRN组,而CRH组和DRH组显著低于CRN组和DRN组。DRN组和DRH组心肌凋亡指数(AI)显著高于CRN组和CRL组,DRL组心肌AI显著高于CRL组。Pearson相关分析显示,在DRN组与DRL组HSP72 mRNA与心肌AI呈显著负相关(P<0.05),而在DRN组与DRH组HSP72 mRNA与心肌AI无相关性。结论:低强度运动可以增加HSP72基因的表达,抑制糖尿病大鼠的心肌凋亡率,对糖尿病心肌病变具有保护作用。

Abstract:
      Objective:To investigate the effects of different intensity exercises on the expression of HSP72 mRNA and myocardial apoptosis in type 2 diabetic rats.Method: Thirty-six male Sprague-Dawley rats were divided into diabetic rats (DFM, n=18) and control rats (CFM, n=18). The diabetic rats were randomly sub-divided into non-exercises group (DRN), high-exercises group (DRH) and low-exercises group (DRL). The control rats were randomly sub-divided into non-exercises group (CRN), high-exercises group (CRH) and low-exercises group (CRL). The rats with exercises training were forced to run on treadmill for 8 weeks. The expression of HSP72 mRNA in cardiac muscle were determined with RT-PCR and myocardial apoptosis were observed with Tunel staining. Result:The expression of HSP72 mRNA of cardiac muscle in diabetic rats were significant lower than that in control rats. The expression of HSP72 mRNA in CRL and DRL groups were significantly higher than that in CRN and DRN groups. The expression of HSP72 mRNA in CRH and DRH groups were significantly lower than that in CRN and DRN groups. Apoptosis index (AI) of cardiac muscles in DRN and DRH groups were significantly higher than that in of CRN and CRL groups. Myocardial AI in DRL groups was significantly higher than that in CRL groups. There was significantly negative correlation between expression of HSP72 mRNA and AI of cardiac muscles in DRL and DRN groups(P<0.05), which was not observed in DRH and DRN groups. Conclusion: Low-intensity exercises can increase expression of HSP72 mRNA and inhibit apoptosis rate of cardiac muscles in diabetic rats, which play an important role of protection from diabetic cardiomyopathy.
