目的:比较督脉取穴法(人中-百会穴)与督脉取穴法联合背俞取穴法(人中-百会穴+肝俞-肾俞穴)在局灶性脑缺血大鼠的治疗效果上是否存在差异。
方法:将健康成年雄性SD大鼠91只随机分为4组:假手术组、模型组、治疗A组(电针人中-百会穴)、治疗B组(电针人中-百会穴+肝俞-肾俞穴),采用线栓法制作大鼠右侧大脑中动脉闭塞模型(MCAO)。电针刺激在造模成功90min后开始,30min/d。分别选取缺血第1、3、7及14天共4个时间点,运用Longa评分对神经功能恢复情况进行评价,RT-PCR及免疫组化测定缺血侧皮质LINGO1 mRNA和蛋白表达。
结果:假手术组无神经功能障碍。治疗A组和治疗B组运动功能恢复情况较模型组明显改善(P<0.05);LINGO1 mRNA和蛋白表达在模型组中缺血第1天时明显增强第3—14天表达逐渐下降,但第14天时仍较假手术组高(P<0.01);治疗A组和治疗B组在第7—14天时其LINGO1 mRNA和蛋白表达均较模型组低(P<0.05),并且,治疗B组在7—14天时的LINGO1蛋白表达较治疗A组低,二者之间差异有显著性意义(P<0.05)。
结论:局灶性脑梗死大鼠缺血侧脑皮质LINGO1 mRNA和蛋白表达明显上调,在脑梗死急性期给予电针刺激后能下调其表达水平,促进轴突再生,加速神经功能恢复;联合取穴法与单一取穴法比较,能取得更好的治疗效果。 |

Abstract
Objective:To investigate the difference between the effect of electroacupuncture(EA)at “Renzhong-Baihui”(DU26-DU20,RB)and“Renzhong-Baihui+Ganshu-Shenshu”(DU26-DU20+BL18-BL23,RB+GS)after focal cerebral infarction in rats.
Method:The healthy adult male SD rats(n=91) were randomly divided into sham-operated group,model group,treatment group A(RB)and treatment group B(RB+GS) and subjected to establish the model of middle cerebral artery occlusion(MCAO)on the right side by nylon monofilament suture. EA was performed at 90 min after establishing of successful model,30min/d. The neural function recovery was evaluated by Longa's score,RT-PCR and immunohistochemistry techniques were used to detect the expressions of LINGO1 mRNA and protein in ischemic cortex on the 1st、3rd、7th and 14th d respectively after ischemia.
Result: The sham-operated group had no neurological dysfunction. Motor function recovered significantly in treatment group A and treatment group B, compared with model group(P<0.05). The expressions of LINGO1 mRNA and protein in model group enhanced obviously on the 1st d,then gradually decreased on the 3rd—14th d,but were still higher than that in sham-operated group on the 14th d(P<0.01). The expressions of LINGO1 mRNA and protein in treatment group A and treatment group B were all lower than that in model group on the 7d—14d(P<0.05);in addition,the expression of LINGO1 protein in treatment group B was lower than that in treatment group A,there was significant statistical difference between two treatment groups(P<0.05).
Conclusion:The expressions of LINGO1 mRNA and protein in ischemic cortex increased significantly. EA might induce its' down-expression after acute cerebral infarction,promote axonal regeneration and accelerate the neural functional recovery. The therapeutic effect of RB+GS group was better than that of RB group. |
