摘要
目的:通过戊四氮(PTZ)癫痫持续状态(SE)大鼠模型观察激光氧液对海马组织内谷氨酸(Glu)和γ-氨基丁酸(GABA)动态变化及细胞形态的影响。
方法:建立28d SD大鼠戊四氮诱导癫痫持续状态模型。随机分为4组:生理盐水(NS)+5%葡萄糖液(GS)组、NS+激光氧液组、PTZ+GS组、PTZ+激光氧液组。选择造模治疗后第 0、3、7、10、14天5个时间点进行观察,采用高效液相法检测海马组织内Glu和GABA含量,HE染色法观察海马区细胞形态变化情况。
结果:PTZ组造模后第3、7、10、14天海马组织Glu含量较NS组明显增高,尤以造模后第3天升高差异具有显著性(P<0.05)。PTZ+激光氧液治疗组在造模后第3、7、10、14天Glu含量较PTZ组下降,尤以造模后第3天差异有显著性(P<0.05),但仍高于NS组。PTZ组造模后第3、7、10、14天海马组织GABA含量较NS组明显降低,尤以造模后第3、7天降低差异具有显著性(P<0.05)。PTZ+激光氧液治疗组在造模后第3、7、10、14天GABA含量较PTZ组升高,尤以造模后第14天升高差异有显著性(P<0.05)。
结论:大鼠癫痫持续状态后一定时间内海马Glu含量升高,而GABA含量降低。激光氧液能够降低大鼠癫痫发作后异常增加的Glu含量,升高GABA的含量,但对GABA含量的影响明显慢于Glu。 |

Abstract
Objective:To study the effects of laser oxygenate fluid on the contents of glutamic acid(Glu) as well as γ-aminobutyric acid(GABA) and the morphology of cells in rats' hippocampus following pentylenetetrazol(PTZ)-induced status epilepticus(SE).
Method:Postnatal day 28(P28) Sprague Dawley(SD) rats received PTZ intra-abdominally to make the model of SE.The rats were divided randomly into PTZ+5% glucose solution(GS) group, PTZ+laser oxygenate fluid group, normal saline (NS)+GS group and NS+laser oxygenate fluid group.Each group was examined on the zero d, 3rd d, 7th d, 10th d and 14th d after SE.The contents of Glu and GABA in homogenized brain tissue of rats were determined with high performance liquid chromatography. The morphology of cells in hippocampus of rats was examined by HE staining.
Result:Contents of Glu was higher on the 3rd d,7th d, 10th d and 14th d,especially higher on the 3rd d after SE developed in PTZ group than that in NS group(P<0.05).The content of Glu in PTZ+ laser oxygenate fluid group was lower than that in PTZ group,especially lower on the 3rd d (P<0.05). Content of GABA decreased after sizure on the 3rd d,7th d,10th d and 14th d,especially lower on the 3rd d, 7th d after SE developed in PTZ group than that in NS group(P<0.05),the content of GABA in PTZ+ laser oxygenate fluid group was higher than that in PTZ group,especially higher on the 14th d(P<0.05).
Conclusion:Content of Glu increased after SE developed for a period of time, while GABA decreased. Laser oxygenate fluid could decrease the unusual increased content of Glu in hippocampus. and increase the content of GABA .But the influence of GABA was obviously slower than that of Glu. |
