      摘要 目的:探讨电针刺激对大鼠早期局灶性脑缺血再灌注损伤细胞凋亡的机制。 方法:选用96只雄性SD大鼠,随机分为假手术组、模型组、电针组各32只。采用改良Longa线栓法制作缺血再灌注大脑中动脉闭塞(MCAO)大鼠模型,电针组于脑缺血再灌注2h后开始电针患侧“曲池”、“足三里”穴。各组于术后24h行神经行为学评分,TUNEL法检测脑组织细胞凋亡,免疫印迹法及逆转录PCR法检测脑组织细胞凋亡相关蛋白Bcl-2,Bax的表达,ELISA检测血清BDNF的变化。 结果:脑缺血再灌注24h后,电针组大鼠神经行为学评分较模型组差异有显著性,且脑组织中TUNEL阳性细胞数明显降低(P<0.05);与模型组相比,电针组Bcl-2表达增高,Bax表达下降;模型组与电针组血清脑源性神经营养因子(BDNF)水平较假手术组相比均明显下降,但电针组较模型组有进一步的升高,差异有显著性意义(P<0.05)。 结论:电针刺激可抑制脑组织中Bax的表达,提高Bcl-2,BDNF的作用,对抗脑缺血再灌注损伤所致的细胞凋亡,达到加速神经功能恢复的目的。

      Abstract Objective: To study the mechanism of anti-apoptotic effect of electroacupuncture (EA) in acute local cerebral ischemia-reperfusion(I/R) rats. Method: Ninety-six male adult SD rats were randomly divided into three groups: sham-operated group, model group and EA group. The model of middle cerebral artery occlusion(MCAO) was established with thread ligation method. The EA group were administered with EA at Quchi (LI11) and Zusanli (ST36) acupoints on the contralateral paralyzed limb at 2h after cerebral I/R. Then, 24h after I/R, rats brains were taken out for TUNEL, Western-blot and RT-PCR examinations, and the blood for ELISA examinations. Result: At 24h after I/R, neurological behavior improved, and the apoptotic cells in ischemic brains decreased in EA group compared with model group (P<0.05). The expression of Bcl-2 increased and Bax decreased significantly in EA group, brain derived neurotrophic factor(BDNF) in blood was milder in EA group than those in model group (P<0.05). Conclusion: EA can significantly regulate anti-apoptotic effect and promote neurological function recovery in cerebral I/R rats in early stage.
