      摘要 目的:探讨Notch信号通路在低声压次声对骨髓间充质干细胞(BMSCs)生物活性影响中的作用。 方法:应用全骨髓培养法获取BMSCs,取第3代对数生长期的细胞,分为四组,分别为第1组(抑制剂+无次声组)、第2组(抑制剂+次声组)、第3组(无抑制剂+无次声组)、第4组(无抑制剂+次声组),按分组要求分别用γ-分泌酶抑制剂DAPT和低声压次声进行相应处理,利用MTT方法检测各组中骨髓间充质干细胞的增殖情况。 结果:低声压次声作用后的24h、48h、72h和96h时,第3组(无抑制剂+无次声组)的OD值比第1组(抑制剂+无次声组)较大,第4组(无抑制剂+次声组)的OD值也大于第2组(抑制剂+次声组),比较均有显著性差异(P<0.05)。另外,低声压次声作用后24h、48h和72h时,第2组的OD值显著大于第1组的OD值(P<0.05);第4组的OD值也显著大于第3组的OD值(P<0.05)。随着作用时间的延长,四组细胞的OD值均呈递增趋势;在次声作用后24h、48h、72h、96h时,第4组的OD值均大于其他三组(4>3>2>1)。 结论:低声压次声可促进BMSCs的增殖活性,Notch信号通路被抑制后会影响低声压次声对BMSCs的增殖作用,说明低声压次声促进骨髓间充质干细胞增殖的机制可能是通过激活Notch信号通路实现。

      Abstract Objective: To investigate the role of Notch signaling pathway in the effect of infrasound on multiplication of bone marrow mesenchymal stem cells (BMSCs). Method: The primary BMSCs were derived from Sprague-Dawley rats through whole bone marrow adherent cultivation. The cells of passage 3 were randomly divided into 4 groups with different treatments as follow: group 1 (DAPT without infrasound), group 2 (DAPT with infrasound), group 3 (without any treatment) and group 4 (infrasound without DAPT). The proliferation of BMSCs in above 4 groups was measured by MTT assay. Result: After being treated with infrasound for 24h, 48h, 72h and 96h, the optical density (OD) value of group 3 is significantly higher than group 1, group 4 significantly higher than group 2 (P<0.05). The OD value of group 2 treated with infrasound for 24h, 48h, 72h is significantly higher than that in group 1, group 4 higher than group 3 (P<0.05). The OD value of four groups gradually increase and group 4 was the highest. Conclusion: Infrasound could promote the proliferation of BMSCs. What's more, such proliferative effects promoted by infrasound could be weakened when Notch signaling pathway was inhibited. It indicated that one of the mechanisms of infrasound promoting proliferation was activation of Notch signaling pathway.
