目的:观察有氧运动对ApoE-/-小鼠动脉粥样硬化(AS)的预防效果,探讨脂联素/AMPK的作用机制。
方法:采用8周龄雄性C57BL/6J和ApoE-/-小鼠进行运动预防AS实验,分为14周对照组(C,10只)、14周模型组(H,10只)、14周运动组(HE,10只)。运动干预结束后取材比较各组小鼠主动脉管壁的病理变化,检测各组小鼠血脂、血糖和血清脂联素,脂肪组织脂联素mRNA表达,以及腓肠肌AdipoR1、AMPK蛋白表达水平。
结果:①C组小鼠体重和脂体比显著低于H组(P<0.01),脂体比HE组小鼠显著低于H组小鼠(P<0.01);②C组小鼠主动脉结构正常,HE组小鼠主动脉管壁纤维弹力板和内膜较H组小鼠完好;③C组小鼠血清TC、TG、FFA及FBG水平均显著低于H组小鼠(P<0.01),HE组小鼠FFA、FBG水平均显著低于H组小鼠(P<0.01);④C组小鼠脂肪组织脂联素mRNA表达及血清水平显著高于H组小鼠(P<0.01),H组小鼠脂肪组织脂联素mRNA表达及血清水平显著低于HE组小鼠(P<0.05);⑤C组小鼠腓肠肌AdipoR1、AMPK蛋白表达均显著高于H组小鼠(P<0.01),HE组小鼠腓肠肌AdipoR1和AMPK蛋白表达均显著高于H组(P<0.05)。
结论:有氧运动具有降低ApoE-/-高脂饮食小鼠AS发生程度的作用,可能与脂肪组织分泌脂联素增多,通过骨骼肌AdipoR1受体/AMPK调节糖脂代谢有关。 |

Abstract
Objective: To observe the prevention effect of the aerobic exercise on ApoE-/- rats AS and to discuss the mechanism of adiponectin/AMPK regulating the glycolipid metabolism in this process.
Method: Ten male C57BL/6J rats and 20 male ApoE-/- rats(8 weeks old) were taken for the experiment. The rats were divided into 14-weeks control group (C, n=10), 14-weeks model group (C, n=10) and 14-weeks exercise group (E, n=10). At the end of the experiment, the pathological changes of the aortic of rats in the three groups were compared and the adiponectin mRNA expression level of adipose tissue, serum adiponectin level, blood lipid, blood glucose and gastrocnemius AdipoR1, AMPK protein expression level were detected.
Result: ① Compared with C group, body weight and perirenal fat were increased significantly in H group, perirenal fat of H group was significantly higher than that of HE group; ② The vascular of C group is normal, fiber elastic plate and internal membrane of the aortic in HE group was better than that of H group; ③ Compared with C group, serum TC, TG, FFA and FBG were lower than that of H group, FFA, FBG of HE group were significantly lower than that of H group (P<0.01); ④ Adiponectin mRNA expression of adipose tissue and serum adiponectin in C group were significantly higher than that of H group (P<0.01), adiponectin mRNA expression of adipose tissue and serum adiponectin in HE group were significantly higher than that of H group (P<0.05); ⑤ Gastrocnemius muscle AdipoR1 and AMPK protein expression in C group were significantly higher than that of H group (P<0.01), gastrocnemius muscle AdipoR1 and AMPK protein expression in HE were significantly higher than that of H group (P<0.05).
Conclusion: The mechanism of prevention ApoE-/- mice AS by aerobic exercise is to activate AMPK through improvement of the serum adiponectin level and gastrocnemius AdipoR1 protein level. |
