      摘要 目的:探讨小鼠骨骼肌挫伤修复过程中肌再生因子与炎性因子的表达规律。 方法:48只雄性ICR小鼠,随机选择8只作为对照组(C组,n=8),其余小鼠以钝物击打构建骨骼肌挫伤模型(S组,n=40),分别在伤后第12h、1天、5天、10天和15天取腓肠肌待测。右侧腓肠肌制作石蜡标本,用于HE染色,观察骨骼肌损伤修复过程中形态变化;左侧腓肠肌用于基因表达分析,荧光定量PCR检测肌再生调节因子与炎性因子mRNA表达变化。 结果:①骨骼肌挫伤后第5天可见少量再生肌纤维,第10天显著增多,第15天基本恢复正常;②骨骼肌损伤后,肌再生因子MyoD、Myogenin、IGF-1、MGF、HGF、TGF-β1mRNA表达量显著增加(P<0.05或P<0.01),GDF-8 mRNA在伤后表达量未出现显著变化;③骨骼肌损伤后,巨噬细胞标志物CD68、CD163、CD206,多种炎性因子IL-1β、IL-6、IL-10和趋化因子CCL2、CCL3、CCL5、CCL8、CXCL10、CXCL12mRNA表达量显著上调,且多在伤后第5天达到峰值(P<0.05或P<0.01)。 结论:趋化因子和炎性因子可能参与了挫伤骨骼肌早期炎症反应,而多种肌再生正向调控因子如MyoD、Myogenin、IGF-1、MGF、HGF可能参与了挫伤骨骼肌修复再生。

A study on the expression of muscle regeneration factor, inflammatory factor in the repair of skeletal muscle contusion in mice    Download Fulltext

Abstract:
      Abstract Objective:To investigate the regulation of muscle regeneration factors and inflammatory factors during the repair of skeletal muscle contusion in mice. Method: Forty eight ICR male mice were divided into control group (C, n=8) and muscle contusion group (S, n=40). Subsequently, their gastrocnemius muscles were harvested at 12 hour,1day, 5day, 10 day, 15 day after injury. Hematoxylin and eosin (HE) stain were used to assess the changes of muscle morphology. In addition, the gene expression of regeneration regulatory factors and inflammatory factors was analyzed by real-time polymerase chain reaction. Result: ①Morphology of skeletal muscles showed signs of regeneration at 5d post injury. The maximum amount of regeneration muscle fibers appeared during 10d post contusion. 15d post-injury morphology of myofibers nearly recovered to normal. ②After skeletal muscle injury, the mRNA expression of myogenic regulatory factors MyoD, Myogenin, IGF-1, MGF,HGF, and TGF-beta 1 was significantly increased (P<0.05 or P<0.01), There was no significant change in the expression of GDF-8 mRNA after injury. ③After skeletal muscle injury, the mRNA expression of macrophage markers CD68, CD163, CD206, inflammatory factors IL-1β, IL-6, IL-10 and chemokines CCL2, CCL3, CCL5, CCL8, CXCL10, CXCL12 were up-regulated (P<0.05 or P<0.01). Conclusion: Chemokines and inflammatory factors may be involved in the early inflammatory response of contusion skeletal muscle confusion, and many positive regulatory factors of muscle regeneration such as MyoD,Myogenin, IGF-1,MGF,HGF, may be involved in the repair and regeneration of contused skeletal muscle.
