      摘要 目的:探讨早期跑步运动是否通过激活内皮型一氧化氮合酶(activating endothelial nitric oxide synthase, eNOS)在大鼠缺血/再灌注(ischemia /reperfusion, I/R)损伤中发挥保护作用。 方法:50只雄性SD大鼠随机分为5组:Sham组(n=10)、I/R组(n=10)、运动+I/R组(n=10)、eNOS抑制剂(L-NIO)+I/R组(n=10)和运动+L-NIO+I/R组(n=10)。I/R大鼠行大脑中动脉阻塞(middle cerebral artery occlusion, MCAO)60mins,再灌2天后,运动+I/R组和运动+L-NIO+I/R组大鼠在跑步机上连续跑5天,每次30mins。再灌7天后,评估各组大鼠神经功能、脑梗死体积,以及梗死周围区和脑血管P-eNOS和P-AMPK蛋白表达。 结果:运动+I/R组神经行为学评分及脑梗死体积明显低于I/R组、L-NIO+I/R组和运动+L-NIO+I/R组(所有P<0.05)。与I/R组、L-NIO+I/R组和运动+L-NIO+I/R组相比,运动+I/R组梗死周围区和脑血管P-eNOS和P-AMPK蛋白水平较高(所有P<0.05)。 结论:早期跑步运动可能通过激活eNOS减轻I/R大鼠神经血管损伤,改善功能预后。

Abstract:
      Absract Objective: To investigate whether early running could protect rats from ischemia /reperfusion(I/R) injury by activating endothelial nitric oxide synthase(eNOS). Method: Fifty male SD rats were randomly divided into five groups: Sham group (n=10), I/R group (n=10), exercise + I/R group(n=10), eNOS inhibitor(L-NIO) + I/R group(n=10) and exercise + L-NIO + I/R group(n=10). I/R rats were subjected to middle cerebral artery occlusion (MCAO) for 60 minutes, and then perfused for 2 days. Rats in exercise + I/R and exercise + L-NIO + I/R group ran continuously on treadmill for 5 days, 30 minutes each time. After 7 days of reperfusion, the neurological score, cerebral infarction volume, P-eNOS and p-AMPK protein expression in ischemic penumbra and cerebral vessels were evaluated. Result: The neurobehavioral score and cerebral infarction volume in exercise + I/R group were significantly lower than those in I/R group, L-NIO + I/R group and exercise + L-NIO + I/R group (all P<0.05). Compared with I/R group, L-NIO + I/R group and exercise + L-NIO + I/R group, P-eNOS and p-AMPK protein levels in ischemic penumbra and cerebral vessels were higher in exercise + I/R group (all P<0.05). Conclusion: Early running may reduce neurovascular injury and improve functional prognosis of I/R rats by activating eNOS.
