      摘要 目的:观察运动对老年大鼠骨髓间充质干细胞自噬及其骨微结构的影响。 方法:6只6月龄大鼠为青年组,饲养6月后取材;12只27月龄大鼠分为运动组、老年组,运动组大鼠给予运动跑台干预8周。干预结束后, ELISA检测血清骨代谢标志物NTX、PINP、BLAP、TRCAP水平;双能X线骨密度仪检测骨密度;骨显微CT检测骨微结构;RT-PCR、 Western Blot检测Beclin1、LC3-Ⅱ、p62 mRNA及蛋白表达情况。 结果:与青年组相比,老年组血清骨形成标志物PINP、BALP水平均降低(P<0.01),左侧股骨骨密度降低(P<0.01),左侧胫骨骨小梁数量(Th.N)及骨小梁体积比(BV/TV)指标均降低(P<0.01),骨髓间充质干细胞自噬体相关mRNA及其蛋白Beclin1、LC3-Ⅱ、p62表达量均上升(P<0.01);与老年组相比,运动组血清骨形成标志物PINP、BALP水平均升高(P<0.01),右侧股骨骨密度(BMD)升高(P<0.01),右侧胫骨骨小梁数量(Th.N)及骨小梁体积比(BV/TV)指标均升高(P<0.01),骨髓间充质干细胞自噬体相关mRNA及其蛋白Beclin1、LC3-Ⅱ表达水平均降低(P<0.01),p62mRNA及其蛋白表达水平有上升趋势(P>0.05)。 结论:通过运动提高骨髓间充质干细胞自噬水平,促进骨髓间充质干细胞向成骨细胞分化,改善老年大鼠骨量、骨微结构,减轻骨质疏松症。

Abstract:
      Abstract Objective: To observe the effects of exercise on autophagy and bone microstructure of bone marrow mesenchymal stem cells in aged rats. Method: Six rats aged 6 months were collected after 6 months of feeding. Twelve 27-month-old rats were divided into exercise group and elderly group. The rats in the exercise group were given exercise treadmill intervention for 8 weeks. After intervention, the levels of serum bone metabolism markers NTX, PINP, BLAP and TRCAP were detected by ELISA. Bone mineral density was measured by Dual Energy X-ray. Bone Micro-CT was used to detect bone microstructure. The mRNA and protein expressions of Beclin1, LC3-Ⅱ and p62 were detected by RT-PCR and Western Blot. Result: Compared with the young group, the serum levels of bone formation markers PINP and BALP were lower in the elderly group (P<0.01), bone mineral density decreased in the left femur (P<0.01), the number of trabecular bone (Th.N) and the volume ratio of trabecular bone (BV/TV) of the left tibia decreased (P<0.01), the expressions of autophagosome related mRNA and its proteins Beclin1, LC3-Ⅱ, p62 increased in bone marrow mesenchymal stem cells (P<0.01). Compared with the elderly group, the serum levels of bone formation markers PINP and BALP(P<0.01), bone mineral density of right femur (P<0.01), the number of trabecular bone (Th.N) and the volume ratio of trabecular bone (BV/TV) of the right tibia increased in the exercise group(P<0.01), the expression levels of autophagosome related mRNA and its proteins Beclin1 and LC3-Ⅱ were decreased in bone marrow mesenchymal stem cells (P<0.01), the expression levels of p62mRNA and protein showed an upward trend(P>0.05). Conclusion: Exercise can improve the autophagy level of bone marrow mesenchymal stem cells, promote the differentiation of bone marrow mesenchymal stem cells into osteoblasts, improve bone mass and bone microstructure of elderly rats, and alleviate osteoporosis.
