      摘要 目的:研究脉冲电磁场对骨质疏松大鼠骨髓间充质干细胞自噬的作用机制。 方法:24只雌性SD大鼠随机分为假手术组(Sham组)、去卵巢骨质疏松模型组(OVX组)、去卵巢骨质疏松模型+脉冲电磁场组(PEMF组),每组8只。OVX组和PEMF组均通过切除双侧卵巢构建骨质疏松动物模型。PEMF组予以脉冲电磁场干预,参数设置为频率8Hz,强度3.82mT,每次干预40min,每日1次,每周5天,共12周。ELISA法检测各组大鼠血清中大鼠雌二醇(estradiol, E2)和骨碱性磷酸酶(bone alkaline phosphatase, BALP)水平,左胫骨行HE染色,右胫骨行Micro-CT骨扫描及骨微结构定量分析,左股骨及第5腰椎行骨密度检测,右股骨骨髓间充质干细胞采用RT-PCR和Western Blot检测Beclin 1、LC3、P62、Runx-2的mRNA和蛋白表达水平。 结果:脉冲电磁场治疗可提升E2和BALP水平,减少骨质流失。Micro-CT骨扫描和HE切片染色表明脉冲电磁场可增加骨小梁数目、骨体积分数,降低骨小梁分离度,改善骨微结构。与OVX组比较,PEMF组的Beclin 1、LC3、P62的mRNA及蛋白表达水平降低,Runx-2的mRNA及蛋白表达水平升高,差异具有显著性意义(P<0.05)。 结论:脉冲电磁场可能通过抑制骨髓间充质干细胞的自噬,促进骨形成,发挥抗骨质疏松的作用。

      Abstract Objective: The aim of this study was to explore the mechanism of pulsed electromagnetic field-induced inhibition of autophagy on bone marrow mesenchymal stem cells in osteoporotic rats. Method: Twenty-four female Sprague-Dawley rats were assigned randomly into sham operation group(Sham group),ovariectomized model group (OVX group),ovariectomized model+pulse electromagnetic field group (PEMF group),8 rats in each group. The bilateral ovariectomized was used to construct an animal model of osteoporosis animal models in OVX group and PEMF group. Rats in the PEMF group were exposed to PEMF (3.8mT,8Hz,40min/day,5days/week,for 12weeks). E2 and BALP were detected by ELISA in each group. The left tibia was stained with HE and the right tibia was scanned by Micro-CT and quantitative analyzed of bone microstructure. The left femur and the fifth lumbar vertebrae were measured by the bone density. RT-PCR and Western blot were used to detect the mRNA levels and the relative expression of Beclin1, LC3, P62 and Runx-2. Result: Pulsed electromagnetic field could significantly increase E2 and BALP levels in serum,increase the bone mineral density in OP rats. Micro-CT detection and HE section staining showed that pulsed electromagnetic field could improve the trabecular bone microscopic parameters. Compared with the OVX group, PEMF group showed the significantly decreased mRNA and protein expression levels of Beclin1, LC3,and P62, and significantly increased Runx-2 expression level(P<0.05). Conclusion: The inhibition of osteoporosis induced by pulsed electromagnetic fields may be achieved through inhibiting the autophagy of bone marrow mesenchymal stem cells and promoting bone formation.
