目的:探讨重复经颅磁刺激(rTMS)对卒中后抑郁(PSD)模型大鼠行为学及海马c-fos、c-jun蛋白表达的影响。
方法:筛选40只健康雄性SD大鼠按随机数字表法分为对照组、假手术组、模型组和治疗组(n=10)。对照组不给予任何干预处理;模型组和治疗组采用大脑中动脉栓塞法(MCAO)和慢性不可预见温和应激法(CUMS)建立PSD模型;假手术组和治疗组给予rTMS治疗;通过蔗糖水消耗实验、旷野实验和水迷宫实验对各组大鼠行为学进行评估;应用免疫组化实验检测各组大鼠海马c-fos、c-jun的阳性细胞灰度值。
结果:①蔗糖水消耗实验:与模型组比较,治疗组蔗糖水消耗量明显增多(P<0.05);②旷野实验:治疗组大鼠水平及垂直得分较模型组有显著提高(P<0.05);③水迷宫实验:治疗组大鼠较模型组学习和记忆能力都有所改善(P<0.05);④免疫组化实验:治疗组大鼠c-fos阳性细胞灰度值高于模型组(P<0.05),各组间c-jun阳性细胞灰度值差异不显著(P>0.05)。
结论:rTMS能够有效改善PSD模型大鼠的抑郁样行为,其机制可能与海马c-fos、c-jun蛋白的表达有关。 |

Abstract
Objective: To investigate the effects of low-frequency repetitive transcranial magnetic stimulation (rTMS) on behavioristics and expression of c-fos and c-jun in hippocampus of rats with post stroke depression (PSD).
Method: Forty male SD rats were randomly and equally divided into four groups(10 in each): the control group, the sham group, the model group and the treatment group. The control group and sham group were given normal care without any intervention while the PSD model group and treatment group were treated with middle cerebral artery occlusion (MCAO) and chronic unpredictable mild stress (CUMS); The sham group and treatment group accepted the rTMS treatment; The sugar water consumption test, the open field test and water maze test were used to verify the effect of rTMS on the rats with PSD; The expression of c-fos and c-jun were detected by immunohistochemistry.
Result: Compared with the model group, the consumption of sugar of the treatment group increased significantly(P<0.05); In the open-field test, the count of horizontal and vertical movement of the treatment group increased significantly (P<0.05); In the water maze test, compared with the model group, learning and memory ability of treatment group improved (P<0.05); the c-fos positive cells grey value of treatment group significantly increased (P<0.05), c-jun positive cells grey value of each group had no significant difference (P>0.05).
Conclusion: rTMS can effectively improve depression-like behavior in rats with PSD. This may be related to the expression of c-fos and c-jun in hippocampus. |
